Driselase Rice Bran Fraction – Adenosine

Driselase fraction contains two main ingredients, so we try to isolated functional ingredients from the Driselase fraction. Here is a figure of the separation of Driselase fraction. We first use silica gel chromatography. And then one of the fraction, F4 fraction was fractionated by HPLC, using reversed-phase column. And also, again, we separated at least 6 fractions from this F4 fraction. So finally, we found fraction 22 has relatively higher activity for blood pressure lowering activity. So in fraction 22, we identified adenosine as a functional ingredient in this fraction. Adenosine is actually, maybe you know the nucleic acid, so this compound is not unique in the rice bran itself. So in that time, I was a little bit disappointment to our research.

But we had decided to continue this research. So when adenosine supplemented to the same animal in this case, chronic administration of adenosine, 10 mg/L of drinking water treatment significantly reduced blood pressure elevation in SHRSP. And mechanism of adenosine treatment. How adenosine reduce blood pressure? We found plasma nitric oxide levels were significantly increased by adenosine treatment. So maybe this nitric oxide maybe occurs in the blood vessel relaxation and also consequently blood pressure is maybe lowering. After three weeks of the feeding of adenosine, we checked several blood parameters. Plasma free fatty acid and glucose and insulin and leptin, these levels were significantly decreased in adenosine administration. On the other hand, plasma adiponectin was increased by adenosine groups.

Adiponectin has improvement effect of lipid and glucose metabolism maybe. And adiponectin is actually one of the major adipo-cytokine And improves insulin resistance and lipid metabolism also.

mRNA expression level of adiponectin in visceral fat was increased by adenosine administration in SHRSP. In vitro experiments, using 3T3-L1 adipocyte. Differentiated 3T3-L1 adipocyte enhanced secretion of adiponectin by adenosine treatment. These results suggested that adenosine enhances plasma adiponectin levels via upregulation of its mRNA expression and/or secretion from adipocyte. Here is the conclusion of adenosine experiment. Administration on adenosine enhances plasma adiponectin levels and improves glucose and lipid metabolism. Adiponectin also enhances nitric oxide production in endothelial cells. So this upregulated nitric oxide reduces blood pressure in hypertension model.

Continuing on explaining Driselase fraction, Professor Shirakawa went further to isolate other ingredients.

The research identified adenosine as the main functional ingredient from Driselase fraction. Adenosine is a nucleic acid and this compound is not unique in the rice bran itself. But further research finds that adenosine reduces blood pressure. Professor Shirakawa will explain this mechanism in Spontaneously Hypertensive Stroke Prone Rat (SHRSP) experiments.
